Viagra: Reborn as Revatio

June 11, 2005 (PRLEAP.COM) Health News
The FDA approved the drug's main ingredient. Revatio will be available in 20 milligram pills. Viagra is sold in doses of 25 milligrams, 50 milligrams and 100 milligrams. Revatio pills are white and round, while Viagra is sold as a blue, diamond-shaped tablet.

The anti-impotence drug Viagra could provide effective treatment for a life-threatening heart and lung condition.

Pulmonary arterial hypertension, a constriction of the vessels that supply blood to the lungs, affects more than 100,000 people worldwide, according to Pfizer. About 95% of patients survived 18 months on Revatio, compared with the 65% survival predicted for that population, says Gary Burgess of Pfizer, Viagra's maker, who directed the research.

Viagra Test

The researchers studied healthy male volunteers who had breathed a low oxygen atmosphere for 30 minutes to induce pulmonary hypertension.

This low oxygen atmosphere caused a 56% increase in pulmonary artery pressure.
When the low oxygen test was repeated - this time after taking Viagra - their pulmonary artery pressure remained almost normal.

Viagra, chemical name Sildenafil, appears to help blood vessels relax and prevent the constriction that normally accompanies lack of oxygen (hypoxia).

Viagra Effect

The effect is based on a biochemical similarity between the penis and the lungs: both contain large amounts of the enzyme phosphodiesterase. It limits erection of the male organ and constricts the blood vessels around the lungs.

Sildenafil in turn blocks the enzyme, thus permitting a sustained erection as well as better circulation in the lung walls, according to the study.

Lung Disorder

Lungs allow us to breath and get oxygen into the bloodstream and the cells of our bodies. During a normal day, we breathe nearly 25,000 times, and take in (or inhale) large amounts of air. The air we take in is mostly oxygen and nitrogen. But air also has things in it that can hurt our lungs – bacteria, viruses, tobacco smoke, car exhaust, and other air pollutants. Disorders that affect the lungs and airways are called lung or pulmonary disorders. Lung disease is not only a killer, it can be life-long. More than 30 million Americans are now living with lung disease.

Because of the different manner in which tissue cells in the same organ system may respond to varying types of exposure to cancer producing agents, several different types of cancer may be found in any organ.

Viagra works for Babies and children having pulmonary hypertension

Pulmonary hypertension is basically high blood pressure in the lungs. It can be caused by a heart defect or for other reasons that are not well explained. The condition can also occur right after heart surgery. In a baby, pulmonary hypertension is virtually a death sentence as the condition makes it very difficult for the heart to pump blood to the lungs. Now, few studies are suggesting babies and children with this disorder may respond to Viagra.

Viagra works by helping the blood vessels dilate or relax. Doctors say, over time, they hope the blood vessels will restructure to a more relaxed state. Mary Taylor, M.D., Pediatric Cardiologist at Vanderbilt Children's Hospital, says, "There are not lot of options for children with high blood pressure in the lungs." She says nitric oxide is one treatment that works in the short-term for these babies, but there is no long-term solution.

Research and Studies

• Recent studies have shown that adults, whose pulmonary hypertension is reduced with nitric oxide, can benefit similarly with oral Viagra. The drug has been used on some babies and children as an off-label treatment. "We have had many, many children, some here and other children all over the country, who have gotten it for pulmonary hypertension and have tolerated it very well," says Dr. Taylor.
• One of the current studies on Viagra in children is being done at 40 sites across the U.S. Part of that study looks at the use of Viagra through an IV for children after heart surgery. Researchers say they will test to see if the blood vessels respond to Viagra in a short time period right after heart surgery.
• Another study in progress is looking at newborns who have persistent pulmonary hypertension. In this study, researchers are trying to determine the appropriate dose of Viagra for the newborns. Dr. Taylor says newborns are particularly at high risk for pulmonary hypertension after heart surgery. She says, "Pulmonary hypertension is one of the major causes of death and complications after cardiac surgery in newborns, thus the importance of finding a treatment."

Doctors hope Viagra may be the answer. “Will the drug work long-term?" Only time will tell.
